RTI-111, also known as dichloropane or O-401, is a synthetic 3-phenyltropane stimulant related to cocaine. It was developed as a research compound for studying monoamine transporters and stimulant behaviour, rather than as a medicine for recreational use. Laboratory data show potent inhibition of dopamine, serotonin and norepinephrine reuptake, while animal studies describe cocaine-like discriminative and reinforcing effects. Human clinical information is extremely limited.

Chemistry and relationship to cocaine

RTI-111 belongs to the phenyltropane family. Its tropane ring and methyl ester resemble the core of cocaine, but the benzoyloxy group is replaced by a 3,4-dichlorophenyl substituent. This structural change produces a distinct transporter-binding profile. The compound is also referred to as dichloropane in chemical and pharmacological literature.

Pharmacology

RTI-111 is a potent inhibitor of all three monoamine transporters. Published transporter-binding data report nanomolar activity at the dopamine transporter (DAT), serotonin transporter (SERT) and norepinephrine transporter (NET), with the strongest measured affinity at DAT. This distinguishes it from cocaine, which has a substantially different balance of transporter activity (Ranaldi and colleagues, 2000).

In rhesus monkeys, RTI-111 produced reinforcing and discriminative-stimulus effects consistent with traditional psychomotor stimulants acting at the dopamine transporter. When administered before methamphetamine, it shifted the methamphetamine dose-response function to the left, indicating an interaction between the two stimulant effects in that experimental model (Ranaldi and colleagues, 2000).

Subjective effects

The PsychonautWiki profile of RTI-111 describes stimulant effects, although human accounts are scarce. Animal findings support cocaine-like stimulant activity, without establishing what the human experience is like, how intense it is or how long it lasts.

There are no controlled human studies defining its subjective effects. Accounts of cocaine or other phenyltropanes are not direct observations of RTI-111.

Toxicity and research limitations

RTI-111 was primarily developed as a pharmacological research tool. The available evidence is dominated by transporter assays and animal behavioural experiments, not clinical toxicology. No well-characterised human case series has established its acute toxic range, metabolism, dependence potential or long-term effects.

Its potent activity at several monoamine transporters and cocaine-like reinforcing effects in animals indicate abuse-related concerns, but they do not provide a human safety profile. The lack of clinical reports should be understood as a gap in evidence, not proof that the compound is safe.

Related compounds

RTI-111 is related to other 3-phenyltropane research compounds and to cocaine. It should not be confused with RTI-113, a separate phenyltropane analogue with its own pharmacological profile.
