1D-AL-LAD

1D-AL-LAD is the name used for a proposed N1-acyl derivative of AL-LAD, in which a 1,2-dimethylcyclobutane-1-carbonyl group is attached to the indole nitrogen. The name has appeared in online product listings, but forensic work has raised a significant identity problem: material sold as 1D-AL-LAD has been found to contain 1T-AL-LAD, a different lysergamide with a thiophene-2-carbonyl group.

Proposed chemical structure

The intended structure of 1D-AL-LAD combines the N6-allyl substitution of AL-LAD with a dimethylcyclobutanecarbonyl group at N1. It is therefore distinct from 1D-LSD, which has the same proposed N1 group on an LSD rather than an AL-LAD scaffold.

The distinction is chemical, not just a matter of naming. Changing the lysergamide core or the N1 acyl group produces a different molecule, and the pharmacology of one cannot be assumed for another.

The 1T-AL-LAD identification issue

In 2024, researchers reported that products marketed as 1D-AL-LAD might instead contain 1-(2-thienoyl)-6-allyl-nor-LSD, abbreviated 1T-AL-LAD. Their analysis followed earlier findings that some products labelled 1D-LSD actually contained 1T-LSD. The authors synthesised and characterised 1T-AL-LAD using NMR, FT-IR, LC-HRMS and GC-MS (Okada and colleagues, 2025).

Japan’s Ministry of Health later listed 1T-AL-LAD as the ingredient in products sold under the name “1D-AL-LAD”. This official notice reinforces the need to verify the actual substance rather than relying on the label printed on a blotter (Japanese Ministry of Health: newly designated controlled substances).

Pharmacology and reported effects

The pharmacology of the proposed 1D-AL-LAD structure has not been established through reliable receptor, metabolism or clinical studies. The 1T-AL-LAD research was analytical and does not provide a human effect profile for 1D-AL-LAD.

For that reason, effects attributed online to “1D-AL-LAD” cannot be assigned confidently to the proposed molecule. If the material was actually 1T-AL-LAD, those descriptions refer to a different structure.

Research limitations

The central uncertainty is the identity of the material being described. Published evidence has confirmed 1T-AL-LAD in products sold as 1D-AL-LAD, while direct confirmation of the proposed dimethylcyclobutanecarbonyl compound in seized retail material has not been established in the sources cited here.

Until an analytically confirmed sample of the intended structure is documented and studied, its biological activity, metabolism, toxicity and human effects remain unknown.

Related lysergamides

1D-AL-LAD is structurally related to AL-LAD, 1cP-AL-LAD and 1D-LSD. It should be distinguished from 1T-AL-LAD, which has a thiophene-carbonyl group rather than the proposed dimethylcyclobutanecarbonyl substituent.
