TMA-6, or 2,4,6-trimethoxyamphetamine, is a synthetic psychedelic amphetamine and a positional isomer of TMA-2. Its three methoxy groups occupy the 2-, 4- and 6-positions of the aromatic ring. It is listed in chemical reference databases and has appeared in research-chemical and drug-checking contexts, but there is very little compound-specific pharmacological or clinical research.
Chemistry and nomenclature
TMA-6 is an alpha-methylated phenethylamine with a 2,4,6-trimethoxy substitution pattern. It should be distinguished from TMA-2, which is 2,4,5-trimethoxyamphetamine, and from TMA, which is 3,4,5-trimethoxyamphetamine.
The naming of trimethoxyamphetamine isomers can be confusing in analytical records. A 2022 CFSRE drug-material report listed several TMA names as synonyms but stated that the sample appeared to contain a 2,4,5-trimethoxy configuration and that the exact positions had not been confirmed by NMR. That sample therefore cannot be treated as definitive analytical confirmation of TMA-6 (CFSRE trimethoxyamphetamine report, 2022).
The US FDA substance database identifies TMA-6 specifically as 2,4,6-trimethoxyamphetamine (FDA Substance Registration System: TMA-6).
Pharmacology
There is little direct receptor research specific to TMA-6. Its classification as a psychedelic is based largely on its structure and historical descriptions of trimethoxyamphetamine compounds. The pharmacology of TMA-2 or 3,4,5-trimethoxyamphetamine should not be transferred directly to TMA-6, since the positions of the methoxy groups can alter receptor interactions.
Comparative research on related trimethoxyamphetamines has demonstrated that positional isomers can differ in their behavioural and receptor profiles. A mouse study of TMA-2 and other mescaline-related compounds found distinct structure–activity relationships between 2,4,5- and 3,4,5-substituted phenylalkylamines (Mescaline analogue head-twitch study, 2019).
Subjective effects
The PsychonautWiki profile of TMA-6 and historical descriptions provide limited information. Controlled human observations are too limited to establish characteristic visual, cognitive or emotional effects.
Although psychedelic activity is plausible from its relationship to other trimethoxyamphetamines, detailed descriptions based on TMA-2 or TMA should not be presented as confirmed effects of TMA-6.
Toxicity and research limitations
No detailed human intoxication series, pharmacokinetic study or toxicological assessment specific to TMA-6 was identified in the available literature. Its acute toxic range, metabolism, dependence potential and long-term effects remain unknown.
Analytical identification is particularly important for this group because several trimethoxyamphetamine isomers have similar names and molecular compositions. A product label alone is not enough to establish which positional isomer is present.
Related compounds
TMA-6 is related to TMA-2 and TMA (3,4,5-trimethoxyamphetamine). These compounds share a trimethoxyamphetamine framework but differ in the positions of the methoxy groups.
