SKU: Flunitrazolam Category:

Flunitrazolam is a synthetic triazolobenzodiazepine related to flunitrazepam and clonazolam. It has never been marketed as a medicine and is known mainly from seizures of designer-drug tablets and forensic toxicology. Although its structure suggests potent benzodiazepine activity, direct information about its effects and safety in humans is limited.

Chemistry and history

Flunitrazolam combines the triazole ring found in compounds such as alprazolam with the nitro- and fluorophenyl-substituted structure of flunitrazepam. It is sometimes described as the triazolo analogue of flunitrazepam. It was first identified in seized tablets in Sweden in 2016 and was subsequently detected by forensic laboratories in Germany and other European countries.

Pharmacology

Flunitrazolam is expected to act as a positive allosteric modulator at the benzodiazepine site of the GABAA receptor. This is inferred from its structural relationship to established benzodiazepines; a detailed receptor-binding study has not established its potency or selectivity. A review of designer benzodiazepines notes that clinical information on flunitrazolam’s effects, safety and tolerability was not available at the time of publication (New/Designer Benzodiazepines: An Analysis of the Literature and Psychonauts’ Trip Reports, 2020).

Metabolism and detection

A study published in 2019 examined flunitrazolam in human liver microsomes and urine. The researchers proposed several metabolites, including desnitro-, 7-amino- and 7-acetamido-flunitrazolam. In the urine sample, the parent compound was detectable for a limited period, while metabolites extended the detection window. The study was based on a single self-administration observation and should not be interpreted as a clinical pharmacokinetic trial (Ameline and colleagues, 2019).

A separate in-vitro study identified phase I metabolites after incubation with pooled human liver microsomes, adding further analytical markers for forensic testing (Moosmann and colleagues, 2019).

Reported effects

Reliable human descriptions of flunitrazolam are scarce. Its expected effects are those associated with benzodiazepine receptor modulation: sedation, reduced anxiety, muscle relaxation and impaired memory. Online reports describe a strong hypnotic effect, but they are not sufficient to establish its relative potency or duration.

Sedation and memory

Because flunitrazolam is structurally related to potent triazolobenzodiazepines, pronounced drowsiness and anterograde amnesia are concerns. However, no controlled human study has quantified these effects for flunitrazolam itself.

Toxicity and research limitations

There is no well-defined human toxic range, and the risk of dependence or withdrawal has not been established specifically for flunitrazolam in the available literature. Its detection in forensic samples confirms non-medical exposure, but does not by itself establish the clinical effects in each case.

As with other benzodiazepines, combining flunitrazolam with opioids, alcohol or other sedatives may produce additive central nervous system depression. This is a general pharmacological concern rather than a quantified interaction profile for flunitrazolam.

Related compounds

Flunitrazolam is related to Flunitrazepam, Clonazolam and Triazolam. Its combination of a triazole ring and flunitrazepam-like substitution distinguishes it from these compounds.
