4′-Fluorococaine is a synthetic cocaine analogue in the tropane family. It differs from cocaine by a fluorine atom at the para position of the benzoyl phenyl ring. The compound has been studied mainly as a radiolabelled research analogue for positron emission tomography (PET), rather than as a well-characterised recreational drug. Its transporter activity differs from cocaine in an important way: laboratory assays found similar potency at the dopamine reuptake site but much stronger activity at the serotonin reuptake site.

Chemistry

4′-Fluorococaine retains the tropane skeleton and methyl ester found in cocaine. The fluorine substituent is attached to the para position of the benzoyl phenyl ring. The prime mark in its name distinguishes this ring position from substitutions on the tropane structure itself.

4′-Fluorococaine is distinct from fluorinated phenyltropanes such as CFT. These compounds share a tropane framework but differ in their ester and benzoyloxy groups, which can substantially change transporter affinity and brain distribution.

Pharmacology

The main compound-specific pharmacology study was conducted in rat brain membrane preparations. It found that cocaine and 4′-fluorococaine were approximately equipotent at the dopamine reuptake site. At the serotonin reuptake site, however, 4′-fluorococaine was about 100 times more potent than cocaine (Gatley and colleagues, 1994).

The study used radiolabelled forms of cocaine and 4′-fluorococaine to examine how cocaine metabolites contribute to PET images of the baboon brain. The similar brain imaging results helped show that labelled metabolites did not substantially distort the distribution pattern observed with cocaine. The work was designed for neuroimaging research, not to establish a recreational or clinical profile.

Research use and occurrence

4′-Fluorococaine has been discussed in the context of cocaine-derived PET tracers and dopamine-transporter imaging. A European national report from 2012 also noted that products marketed as 4-fluorococaine were being offered online as a legal alternative to cocaine. That observation documents its appearance in the early research-chemical market, but does not establish widespread use or the identity of products sold under that name.

Subjective effects and toxicity

There is no reliable clinical literature defining the subjective effects of 4′-fluorococaine in humans. The transporter findings suggest that its activity may not simply reproduce the effects of cocaine, particularly because of its much stronger serotonin-reuptake activity in the reported assay. However, the rat membrane results cannot predict the character or intensity of a human experience.

Its acute toxicity, metabolism after non-radiolabelled exposure, dependence potential and long-term effects have not been adequately characterised. It should not be assigned a standard cocaine-like effect profile solely on the basis of structural similarity.

Related compounds

4′-Fluorococaine is related to Cocaine and other substituted tropane compounds investigated as monoamine-transporter ligands. It is chemically distinct from synthetic cathinones, despite sometimes appearing in broad lists of research stimulants.
