Ephylone, also known as N-ethylpentylone, bk-EBDP or βk-EBDP, is a synthetic cathinone stimulant with entactogenic properties. It is structurally related to pentylone and ethylone, but has a longer alpha-carbon chain than ethylone and an N-ethyl group. Ephylone became prominent in the illicit drug market in 2016–2017 and has been linked to numerous severe intoxications and deaths.

Chemistry and history

Ephylone contains a methylenedioxy-substituted phenyl ring, a β-keto group, an N-ethylamino group and a propyl substituent at the alpha carbon. It is the N-ethyl analogue of pentylone. The compound was first detected in drug seizures in 2016 and became particularly common in forensic casework in the United States during 2017.

It has also been found in products sold as MDMA or other stimulant-entactogens, which complicates the interpretation of reported effects and intoxications.

Pharmacology

Ephylone acts as a potent inhibitor of the dopamine, norepinephrine and serotonin transporters. In vitro studies found strong activity at DAT and NET, with weaker activity at SERT. Unlike some MDMA-like cathinones, it did not show transporter-mediated monoamine release in the study that characterised its mechanism. The resulting profile is consistent with a potent psychomotor stimulant (Ephylone pharmacology and clinical case series, 2018).

Subjective effects

The PsychonautWiki profile of ephylone draws on anecdotal accounts of stimulant and entactogenic effects. Users mention euphoria, increased energy and sociability, alongside a strong tendency towards agitation and compulsive redosing. These reports cannot predict an individual experience.

Physical effects

Reported effects include stimulation, increased heart rate, sweating, palpitations and raised body temperature. Some accounts describe tremors, jaw tension, dehydration and reduced appetite. Severe intoxication reports include hyperthermia, rhabdomyolysis and cardiovascular collapse.

Cognitive and psychological effects

Users describe euphoria, increased alertness and faster thinking. At higher levels of intoxication, agitation, confusion, paranoia, hallucinations and delusional thinking have been reported. Some cases involve psychosis and aggressive behaviour.

Clinical and forensic findings

A clinical case series found ephylone in blood samples from intoxicated patients, with concentrations ranging from 7 to 170 ng/mL. Reported presentations included palpitations, tachycardia, agitation, hallucinations, coma and death. The authors concluded that ephylone can cause serious cardiovascular and neurological adverse effects (Ephylone pharmacology and clinical case series, 2018).

A review of synthetic cathinones reported both fatal and non-fatal ephylone poisonings. The clinical findings included cardiac arrest, hyperthermia, rhabdomyolysis, tachycardia, psychomotor agitation, psychosis and renal complications. The wide range of concentrations in forensic samples reflects the complexity of mixed-drug cases and post-mortem interpretation (Review of synthetic cathinones, 2019–2022).

Toxicity and research limitations

There is substantial evidence that ephylone can be associated with severe acute toxicity. Animal studies have also found rewarding and reinforcing effects, as well as neurotoxic and cardiotoxic signals in experimental models. These findings raise concern about abuse liability, but they do not establish the long-term consequences of repeated human exposure.

Many fatal cases involved additional substances, and the contribution of ephylone must be interpreted in that context. Nevertheless, the number and severity of reported cases make it one of the more concerning cathinones in forensic surveillance.

Related compounds

Ephylone is related to Butylone, Ethylone and Pentylone. It is the N-ethyl analogue of pentylone and should not be confused with ethylone, which has a shorter alpha-carbon chain.
