Methylone, also known as 3,4-methylenedioxy-N-methylcathinone (bk-MDMA), is a synthetic cathinone with stimulant and entactogenic effects. It is the beta-keto analogue of MDMA and was first synthesised in the 1990s. Methylone appeared on recreational drug markets in the 2000s and has been sold as a substitute for MDMA. Controlled human studies show that it can produce euphoria, stimulation and increased empathy, alongside cardiovascular effects.

Chemistry and history

Methylone contains a methylenedioxy-substituted phenyl ring, a beta-keto group and an N-methylamine side chain. Its structure differs from MDMA by the addition of a carbonyl group at the beta position. It is closely related to Ethylone and Butylone, which have different nitrogen or alpha-carbon substitutions.

The compound was first described in patent literature in 1996. It later appeared on the recreational market as a product sold under names such as Explosion and was sometimes marketed as an alternative to MDMA.

Pharmacology

Methylone acts as a substrate-type releaser at dopamine, norepinephrine and serotonin transporters. It enters nerve terminals through monoamine transporters and promotes neurotransmitter release. Its profile is similar to MDMA, although differences in potency and transporter selectivity affect the balance of its stimulant and entactogenic effects.

In a controlled study comparing methylone with MDMA in recreational users, methylone increased blood pressure and heart rate and produced stimulation, euphoria, well-being, enhanced empathy and altered perception. Its overall profile resembled MDMA, but the onset was faster and the subjective effects faded earlier. The findings suggested a similar abuse liability under the conditions studied (Poyatos and colleagues, 2021).

Subjective effects

The PsychonautWiki profile of methylone includes users’ comparisons with MDMA, describing stimulation and entactogenic effects. The emotional effects are frequently described as shorter-lived or less pronounced.

Physical effects

Stimulation, increased energy and physical euphoria are commonly reported. Other effects include increased heart rate, elevated blood pressure, sweating, jaw tension, reduced appetite and difficulty sleeping. Some users describe nausea, dehydration and headaches.

Emotional and cognitive effects

Euphoria, empathy, sociability and increased appreciation of music are recurring descriptions. Users may report emotional openness and disinhibition, together with faster thoughts and increased alertness. Anxiety, agitation and confusion can occur, particularly when the effects become uncomfortable or prolonged.

Clinical toxicity

Clinical reports describe tachycardia, hypertension, hyperthermia, agitation, seizures, hallucinations and psychosis. Severe intoxications may involve cardiovascular complications and altered consciousness. Methylone is often detected with other substances, so individual case reports do not always establish its independent contribution.

The controlled human study is useful because it confirms cardiovascular and subjective effects under monitored conditions, but it was not designed to assess rare severe outcomes or repeated exposure.

Metabolism and longer-term effects

Metabolism includes demethylenation, O-methylation, N-demethylation and beta-ketone reduction. A 2025 study of methylenedioxy-substituted cathinones identified several phase I and phase II pathways for methylone and related compounds, providing markers for forensic detection (Yeh and colleagues, 2025).

Long-term effects and dependence potential have not been fully established. Evidence from MDMA or other cathinones can provide context, but does not replace compound-specific studies of methylone.

Related compounds

Methylone is related to Ethylone, Butylone, Eutylone and MDMA. It is the beta-keto analogue of MDMA and the N-methyl counterpart of ethylone.
