2-NMC is a synthetic cathinone identified in analytical research on emerging psychoactive substances. The abbreviation is used for 1-(4-methylphenyl)-2-(dimethylamino)propan-1-one, a dimethylated cathinone with a methyl group at the para position of the phenyl ring. It has been investigated in an in-vitro metabolism study, but there is little published information about its pharmacological effects or human toxicity.
Chemistry
2-NMC contains the β-keto phenethylamine framework characteristic of cathinones. Its structure has a 4-methyl-substituted phenyl ring and a dimethylamino group on the alpha carbon. The nitrogen substitution distinguishes it from more familiar N-methyl cathinones such as 3-MMC and 4-MMC.
The abbreviation should not be confused with 2-MMC, which is 2-methylmethcathinone. Despite the similar names, the two compounds have different structures: 2-MMC has an ortho-methyl group on the ring and a secondary N-methylamine, whereas the 2-NMC described in the analytical literature has a para-methyl group and a dimethylamine.
Pharmacology
There is no well-established receptor or transporter profile for 2-NMC. Its classification as a synthetic cathinone follows from its chemical structure, but the stimulant activity of related cathinones is not enough to establish its potency or precise mechanism.
A 2022 study examined the metabolism of 2-NMC alongside three other synthetic cathinones. Using human liver microsomes, the researchers identified phase I and phase II metabolites and reported 24 metabolites across the compounds studied. The work provides analytical markers that may help identify exposure in toxicology samples, but it does not establish the subjective effects or clinical toxicity of 2-NMC (Gavrilović and colleagues, 2022).
Reported effects
Reliable human accounts of 2-NMC are too scarce to establish a characteristic experience. No controlled study has established whether it produces the same balance of stimulation, euphoria or cardiovascular effects as better-known cathinones. A detailed description of its subjective effects would therefore rely on speculation.
Metabolism and toxicology
The available metabolic research shows that 2-NMC undergoes biotransformation in human liver microsomes, but its full metabolic pathway and the significance of individual metabolites have not been established in clinical samples. There is no published human pharmacokinetic study defining its duration or elimination.
No reliable clinical series has characterised acute poisoning, dependence or long-term effects. The lack of case reports should not be interpreted as evidence of low toxicity; it reflects the limited amount of compound-specific research.
Related compounds
2-NMC is related to other synthetic cathinones, including 3-MMC, 4-CMC and 4-EMC, but its dimethylated nitrogen distinguishes it from many common N-methyl and N-ethyl cathinones.
